Question about axle gearing and locker.

Can I use the same locker if I upgrade a Dana 44 with 373 gears to 456 gears or will I need a new locker?

That depends on the locker. You wouldn't need a new lunchbox locker as they don't care what ratio carrier they're installed into, but I never recommend a lunchbox locker for the rear axle.

But for a full carrier locker like a Detroit Locker, the carrier ratio break is 3.73 and higher, or 3.92 and lower. So you'd either need a new locker when converting from 3.73 to 4.56, or you'd need to use a 'thick' style R&P gear set to continue using the locker installed when you had the 3.73 gearing. The 'thick' refers to the ring gear being made thicker to allow its use on a carrier that wouldn't normally allow a 4.56 or lower ratio.
